TROPLIST: in situ followed by antibody staining

Hi all,
We are interested in performing in situs followed by antibody
staining with a myc antibody and are seeking advice on the best way
to do this. We will be following the standard Harland lab/Sive
protocol for the in situ. What is the best way to stop the reaction
and perform the post-fix for the transition to the myc staining? We
are thinking to stop the BCIP/NBT reaction with PBT and then fix with
MEMFA for 20 minutes, followed by block and anti-myc incubation. We
will be using a DAB reaction to detect the anti-myc. Since these are
trop embryos, we will probably want to bleach them, so we are
planning to fix with Bouin's at this point, followed by xylene,
paraplast embedding and sectioning.
Any comments or suggestions?
